In qPCR, a probe is a short, labeled DNA or RNA sequence that binds specifically to the target DNA during amplification. Used alongside primers, probes enable real-time monitoring through fluorescence detection. Each probe carries a fluorescent reporter dye and a quencher; when the probe is cleaved during amplification, the reporter is released, causing an increase in fluorescence that reflects the amount of target DNA present.
